What is it for you?
I was definitely disappointed in Iron Man 2 -- I do not believe it was nearly as crappy as some superhero movies I've seen, but it was definitely the biggest letdown compared to my expectations for it.
But for me, the prize goes to Superman Returns, a cinemative experiences that literally does not cover any new ground for a Superman movie, and a movie in which Superman does not punch anything. I remember trying to describe the plot to someone and they replied, "Didn't they already do all that in another movie?" Yeah, pretty much. Ugh.

Iron Man 2 is around the level of Iron Man 1 for me in terms of quality, without being as much of a surprise which really puts it on poor footing compared with the original. Rumors going around say that Favreau was really restrained by the requirements of the Marvel movie shared universe, which probably makes Iron Man 2 the most authentic superhero comic movie ever.
Superman Mopes Returns has so much wrong with it I find it hard to catalog it all. For one film, look at the palette of the whole thing: it's designed to match Superman's costume, which is kind of clever, but they made his costume maroon, teal, and sepia, which isn't clever at all. It is the most dreary looking movie I've ever seen about a man who's supposed to be wearing bright red and blue.
Also, closed captioning actually made a bad movie worse for me retroactively. There's a shout as Luthor's goons are beating the crap out of Superman, and I thought it was one of them. When you put the captions on you realize this is Brandon Routh squealing "I'M STILL SUPERMAN!" Which just seems completely out of character and pathetic to me.

Spider-Man 3, definitely. I'm not so rabid now, but Venom used to be my favorite all-time comics supervillain. To see him reduced to a rather meh appearance at the end of the movie... and to see him so blithely killed... sad.

Does the Ang Lee Hulk movie not exist so much that it can't be mentioned in this thread? I actually enjoyed that one until near the end. The Hulkish dogs and the bizarre final battle, uh, kinda threw the whole thing into chaos.
